AutoPhagyGO Inc. (Headquarters: Suita City, Osaka Prefecture; President and CEO: Miwako Ishido; hereinafter 'AutoPhagyGO') announced the results of the XPRIZE HEALTHSPAN semifinal trial at the 26th Annual Meeting of the Japanese Society of Anti-Aging Medicine held on June 27, 2026. In this study, an 8-week multifaceted intervention combining nutrition, exercise, and sleep targeting autophagy activation was conducted on healthy middle-aged and older adults. The results showed favorable changes in multiple healthspan-related indicators, as well as findings suggesting the utility of human autophagy activity assessment—a method not yet established globally. AutoPhagyGO is currently advancing research and development as one of the 'Top 40' teams selected from global applicants in the international competition 'XPRIZE HEALTHSPAN'. [Key Points of the Presentation] ■ An 8-week intervention trial comprehensively evaluating healthspan in healthy middle-aged and older adults An 8-week program combining autophagy-activating supplements, dietary management, exercise, and sleep management was conducted on healthy individuals aged 50–80, with assessments of physical function, cognitive function, immune function, and other parameters. ■ Favorable changes observed in healthspan-related indicators The 8-week autophagy program was safely implemented with high adherence, resulting in favorable changes in multiple healthspan-related indicators including lower-limb muscle function, muscle quality, cognitive function, fatigue, and sleep. No significant safety concerns were observed. ■ Accumulation of insights toward practical human autophagy activity assessment Results suggestive of autophagy activation were obtained, including increased platelet LC3-II flux and decreased serum and urinary Rubicon levels. Quantitative evaluation of autophagy in humans has not yet been established globally, and this study contributes foundational insights toward its practical application. However, this tri
